Protectie Effects of the Thromboxane A2-Receptor Antagonist, KT2-962 against U 46619-Induced Sudden Death in Cats



Abstract
Thromboxane A2(TXA2) has been implicated in the pathophysiology of cardiovascular disorders and suggests that TX receptor antagonists are effective in cardiovascular disorders.
The purpose of this study was to evaluate the protective effects of KT2-962, an azulene derivative of recently synthesized selective TXA2 receptor antgaonist, against U 46619-induced sudden death in cats. Male cats weighing 3 kg were anesthetized
with
sodium pentobarbital (40mg/kg. I.P). the cats were divided into two groups, U 46619 (100ug/kg, I.V.)-administration group and KT2-962 (5mg/kg, I.V.)-administration group at time of 30 min before the administration of U 46619( 100ug/kg, I.V.) The
cats
were placed on a operating table, and the right carotid artery and cubital vein were cannulated for the recording of mean arterial blood pressure and the injection of drugs. Blood pressure and electrocardiogram were cotinuously monitored on a
physiograph 15 min, 20 min, 25 min and 30 min were measured with blood cell counter (CELL-DYN 1600). The point of death was determined by monitoring blood pressure and electrocardiogram.
@ES the results obtained can be summarized as follows:
@EN 1. In U 46619-administration group. the cat died suddenly between 3 and 5 min and at 5 min after the administration of U 46619, the platelet count was reduced to 62% of the value of time of o min.
2. In KT2-962-administration group at time of 30 min before the administration of U 46619, the cat survied.
These results suggest that KT2-962 may have the protective effects against U 46619-induced sudden death in cats.
